Insilico's AI-designed Pan-TEAD inhibitor ISM6331 hits 40% ORR in mesothelioma Phase 1

Insilico Medicine presented first-in-human Phase 1 data at ESMO 2026 for ISM6331, an oral pan-TEAD inhibitor designed with its Chemistry42 generative AI platform. In heavily pretreated mesothelioma, the 130 mg once-daily dose achieved a 40.0% ORR (2/5 response-evaluable patients, target-lesion shrinkage of 58% and 36%); across 30–130 mg cohorts ORR was 18.2% with 68.2% disease control. Responses appeared by the first Week-8 scan in 3 of 4 responders, one partial response has lasted about a year, and dose escalation continues (NCT06566079, N=36). A notable proof point that generative AI-designed drugs are working in real patients.
