multiDIA Preprint Quantifies All 140 Residues of α-synuclein Without Fractionation

A new bioRxiv preprint from Nikolai Slavov's lab introduces multiDIA, which combines multi-protease independent digestions (trypsin, Lys-C, Glu-C) with data-independent acquisition to boost sequence coverage without chromatographic fractionation. It quantified all 140 residues of α-synuclein across 12 brain donors — including the 45-residue acidic C-terminal tail that trypsin or Lys-C alone cover by only 2 residues. In Parkinson's donors, N-terminal peptides fall below controls, consistent with N-terminal truncation; 3 of 6 PD donors show residues 1–60 averaging 2-fold below control levels.
