A Gladstone Institutes preprint, 'A Compact Regulatory Logic Governs Fibroblast Cell State Plasticity in Fibrosis,' integrates mouse and human epigenomic data (accessibility, H3K27Ac, BRD4, nascent transcription) to identify 280 disease-activated, human-conserved fibroblast enhancers. The team perturbed them via CRISPRi CROP-seq across 549,767 cells and validated enhancer activity with MPRA, using fibroblasts from healthy donors and heart failure/MI patients.
