Anthropic x Adaptyv protein design challenge 2 targets TNF-α with pH 6.0 release twist

Anthropic and Adaptyv announced Challenge 2 of their protein design competition, targeting TNF-α — the inflammation marker blocked by Humira, the best-selling drug ever, and overexpressed in rheumatoid arthritis and Crohn's disease.
The goal is a better TNF-α binder that releases selectively at pH 6.0 in the endosome to improve drug half-life. Designs must also bind both human and mouse TNF-α, enabling the same design to be tested in preclinical models. Organizers note the target is already tough for binder design, and the pH requirement raises difficulty further.
