The CASCADE framework asks four questions of each fine-mapped variant: whether it lies in a peak, changes accessibility, links to a gene, and changes that gene's expression.
Only 3% of variants complete the full cascade, yet these colocalize with disease GWAS signals 10% of the time — roughly twice the rate of chromatin-only variants (3.7–5.4%).
