Sculpta demos bioorthogonal barcoding with >99% accuracy for RNA-seq
anshulkundaje · x · 2026-09-25
Sculpta released the first demo of bioorthogonal barcoding ("bobcoding"): covalently attaching oligo barcodes at multiple internal RNA positions via two-step chemistry, then multiplexed reverse transcription to generate barcoded cDNA libraries with >99% accuracy — claimed to be the first multiplexing before any enzymatic library-prep step, adding one barcode every 300 bases vs. 0-1 for prior methods. Researchers note this could improve many RNA-seq workflows, relevant to "virtual cell" models built on scRNA-seq data with hundreds to thousands of transcripts per cell.
