OptiPrime in Nature Biotech: ML Model Accurately Predicts Prime Editing Efficiencies

David R. Liu's team published OptiPrime in Nature Biotechnology. It is a mechanism-based machine learning model designed to predict prime editing (PE) efficiencies and suggest optimal pegRNAs.
The model incorporates biochemical knowledge and was trained on 300,000 PE efficiencies collected across 40 experimental conditions from multiple labs. OptiPrime successfully identified efficient PE strategies for various in vitro and in vivo applications and could predict outcomes for variants not seen during training (like twinPE), significantly outperforming months or years of manual experimental optimization.
