AIR-seq measures RNA synthesis and decay without chemical conversion

Steady-state RNA-seq only shows how much RNA is present, not the synthesis and decay rates that shape gene expression. This paper introduces analog intrinsic recoding sequencing (AIR-seq), which repurposes N4-hydroxycytidine (NHC) to mark newly synthesized RNA through C-to-T and T-to-C mismatches in standard RNA-seq libraries.
