TWAS false sign rates hit 33% for complex traits, Nature Genetics study finds
zakkohane · x · 2026-10-25
A Nature Genetics study tests how accurately TWAS recover the direction of gene effects on traits. Leveraging the expectation that higher expression should raise protein levels, plus loss-of-function burden tests for complex traits, the authors find false sign rates of 23% for plasma proteins and 33% for complex traits across 484 gene-trait pairs, attributing much discordance to eQTL confounding.
