Landmark study of 12M single cells links T cell ratio to immune aging and mortality

- Unprecedented scale: single-cell RNA-seq across 8 international cohorts, 2,609 individuals aged 18–98, totaling 12,394,834 white blood cells harmonized into 59 immune cell types.
- Core finding: as the thymus involutes, naive T cells ("rookies") decline and are replaced by functionally divergent memory T cell subtypes — GZMK+ vs GZMB+ Tem cells carrying distinct granules. Loss of naive T cells/RTEs was the strongest predictor of chronological age.
- Practical output: the GZMK+/GZMB+ Tem ratio yields a practical blood test. The plotted survival curve shows 31% all-cause mortality over 16 years for high %TemB+ vs 18% for low (p<0.0001).
