Anthropic x Adaptyv's Protein Design Challenge 2 targets TNF-alpha with pH-switchable binders, deadline Oct 11

- The Anthropic x Adaptyv Protein Design Competition has announced Challenge 2: designing binders against TNF-alpha, the inflammation marker protein overexpressed in rheumatoid arthritis and Crohn's disease—best known as Humira's target, the best-selling drug ever.
- The challenge requires a binder that binds at pH 7.4 but selectively releases at pH 6.0 in the cellular endosome, improving drug half-life through a "recycling" mechanism.
- Binders must also be cross-specific to both human and mouse TNF-alpha, enabling preclinical testing in mouse models.
- Deadline: October 11th, 23:59 AoE. Last week's challenge targeted EGFR.
