Nature cover: 6.3M nuclei sequenced to build largest human prefrontal cortex cell atlas

The PsychAD consortium published a Nature cover study building one of the largest single-cell transcriptome atlases of the dorsolateral prefrontal cortex (DLPFC), the region behind working memory, decision-making, and executive function.
- Single-cell RNA sequencing of over 6.3 million nuclei from nearly 1,500 brain donors.
- Over 30% of the 1,494 donors are of non-European ancestry and 21% have two or more major brain diseases, preserving age, genetic background, comorbidity, and clinical phenotype dimensions.
- Across 8 diseases mapped together, individual differences alone explain about 10% of overall transcriptome variation.
- In 284 neurotypical donors aged 0–97 (1.3M+ nuclei), most developmental differences were analyzed (post truncated).
