Topos Bio launches Topos-2 and Topos-Bind to model disordered proteins AlphaFold can't

Post-AlphaFold, structure prediction is treated as solved—but 3 in 5 human proteins have long disordered regions with no single structure. Topos Bio announced Topos-2, an all-atom generative model producing the full conformational ensemble across the order-disorder spectrum, plus a preview of Topos-Bind.
- Topos-2 matches SAXS and NMR experiments more closely than any other model on the independent PeptoneBench benchmark.
- Topos-Bind is claimed to be the first generative model capturing disordered proteins bound to small molecules; on Alzheimer's target amyloid-β with 22 small molecules, it reproduces the broad shape distribution seen in all-atom MD, while cofolding competitors collapse the peptide into a narrow range.
These intrinsically disordered proteins drive signaling, transcription and condensate formation, and are implicated in cancer and neurodegeneration—long deemed "undruggable".
