Michael Levin and Hananel Hazan released a preprint, "Computational Framework for Identifying Ion Channel Mutation-Compensating Interventions," presenting an automated pipeline for finding therapeutic interventions in channelopathies.
The core idea: fix a hardware problem (an ion channel mutation) in software — instead of repairing the mutated channel, compensate for its effects on neural firing patterns by modulating other pharmacologically accessible conductances.
The pipeline:
- Starts from patch clamp recordings of the variant channel
- Searches over pharmacologically accessible conductances
- Outputs which currents must change and by how much
- Guides discovery of electroceutical drugs that open/close other channels to compensate
