Prime assembly: Nature paper shows CRISPR-based in-cell integration of gene-sized DNA fragments

A team led by Daniel E. Bauer published an open-access Nature paper describing prime assembly, a Gibson-cloning-inspired method that leverages CRISPR-targeted dual flap synthesis to assemble and integrate gene-sized DNA fragments directly in human cells, including targeted genomic rearrangements.

The work tackles a core challenge in therapeutic genome editing: installing medium-to-large genomic modifications in therapeutically relevant cells, with potential implications for treating inherited and acquired disorders.
