MIT's PottsMPNN Framework Advances Protein Design Beyond Natural Sequences

MIT's Department of Biology developed PottsMPNN, a machine learning framework to improve protein design. Unlike traditional methods limited to mimicking natural sequences, PottsMPNN incorporates physical principles of structure and stability. It uses noise for diversity and pairwise distributions to capture amino acid interactions. Better at understanding the sequence-energy landscape, it enables the design of structurally feasible, novel proteins without relying on native sequences.
