Moderna Cancer Vaccine Phase 3 Success: AI-Powered Neoantigens
BoWang87 · x · 2026-08-20
Moderna and Merck announced positive Phase 3 results for their individualized mRNA cancer vaccine (INTerpath-001) in resected melanoma (1,137 patients). This is the first successful Phase 3 trial for individualized neoantigen therapy.
Pipeline:
- Surgical resection.
- Whole exome + RNA sequencing.
- ML algorithm identifies somatic mutations and predicts which are immunogenic (displayed on cell surface to trigger T-cell response).
- mRNA encoding up to 34 patient-specific targets.
- Manufactured and shipped in 8 weeks.
Key Validations:
- ML Prediction Works: Tumor-specific neoantigen prediction validated in a large-scale blinded trial.
- Manufacturing Feasibility: 8-week personalized mRNA manufacturing is operationally real.
- Additive Effect: Combination with PD-1 blockade (Keytruda) showed additive benefit, not redundancy.
